coronary arteries, -4 or that they diminish the oxygen requirements of the myocardium by diminishing the external work of the heart.5'6 Much of the confusion concerning the actions of these drugs has arisen from the assumption that nitroglycerin and amyl nitrite have similar effects. Moreover, many studies on the peripheral circulatory effects of nitrites have been carried out with sodium nitrite, administered orally or intravenously, and it has generally been considered that the actions of this substance resemble those of nitroglycerin and of amyl nitrite. In order to determine whether the peripheral circulatory actions of nitroglycerin and amyl nitrite might play a role in their therapeutic effects, the actions of these drugs on both the resistance and capacitance vessels of the forearm of normal human subjects were determined, and the results of these studies are described in this report. The effects of nitroglycerin on ventricular dimensions are described elsewhere.7 The results of these two investigations suggest the manner in which the actions of these drugs on the peripheral circulation affect the heart, and they afford additional insight into the mechanism by which nitroglycerin and amyl nitrite provide relief of anginal pain.

The kinetic differences between sodium nitrite, amyl nitrite and nitroglycerin oxidation of hemoglobin.

The effect of sodium nitrite, amyl nitrite and nitroglycerin (glyceryl trinitrate) on the hemoglobin of adult erythrocytes was examined in vitro. Both amyl nitrite and nitroglycerin reacted immediately with oxyhemoglobin to effect oxidation into methemoglobin while sodium nitrite required an inductionary period (lag phase) prior to the reaction. Kinetic studies of the biomolecular rate law for ...

Left ventricular volume changes after amyl nitrite and nitroglycerin in man as measured by ultrasound.

SUMMARY The therapeutic effect of nitrites in myocardial ischemia is considered to be due in part to a reduction of left ventricular volume but measurement of this parameter in man has been difficult. The technique of echocardiography was used to measure changes in left ventricular end-diastolic volume (LVEDV) and end-systolic volume (LVESV) after administration of amyl ni

Effects of amyl nitrite in aortic valvular and muscular subaortic stenosis.

INHALATION of amyl nitrite causes abrupt vasodilatation and increase in cardiac output and intensifies the murmur of aortic stenosis.1 The transaortic systolic pressure gradient increases because of the increased ejection rate through the stenotic orifice. In muscular subaortic stenosis this effect is particularly marked,2-6 not only because of the increased ejection rate but also because the d...

Influence of Right Ventricular Hemodynamics on Left Ventricular Diastolic Pressure-Volume Relations in Man

To clarify the mechanism of displacement of the left ventricular diastolic pressure-volume function with alteration of loading conditions, the effects of nitroglycerin on pressure-volume relations in 13 patients were compared with those of amyl nitrite in 13 other patients during cardiac catheterization. After nitroglycerin, average systemic mean arterial pressure declined by 15.1 mm Hg (17%) a...

Actions of the nitrites on the peripheral circulation and myocardial oxygen consumption: significance in the relief of angina pectoris.

Myocardial ischemia is best viewed as a discrepancy between myocardial oxygen demands and availability of oxygen to the heart. Although the fundamental action of each nitrite is generalized direct v d l a t i o n , in patients with coronary arterial disease the dynamic factor mediating favorable effects of these agents on angina pectoris appears largely to be reduction of myocardial oxygen requ...
